Flow Cytometry and High-Content Imaging to Identify Markers of Monocyte-Macrophage Differentiation

چکیده
The identification and characterization of cell surface markers present on different subtypes of cells of the immune system can broaden the understanding of immune cell function. The BD LyoplateTM human cell surface marker screening panel is a collection of cell surface marker antibodies designed for screening cells for the expression of 242 human cell surface proteins by flow cytometry or fluorescence imaging. Using a human monocytic cell line, THP-1, undifferentiated (suspension) THP-1 cells were screened for surface marker expression using a BD FACSCantoTM II flow cytometer. Differentiated (adherent) THP-1 macrophages were screened using a BD PathwayTM high-content imaging system. The analysis of results from both proof-of-principle screening experiments identified 21 markers expressed on both THP-1 monocytes and macrophages, 23 markers uniquely expressed on THP-1 monocytes, and 20 markers uniquely expressed on THP-1 macrophages.

Differentiation and Apoptosis of U937 Leukemia Cells by an Active Compound from Dendrostellera lessertii
Background: Regarding the strong differentiation and anti-leukemic activity of Dendrostellera lessertii extract, an active agent with similar capabilities was isolated from the crude extract of the plant leaves. The aim of this study was to determine whether the anti-proliferative effect observed for this compound, is differentiation-dependent among the drug-treated cells, using U937 cells. Me...

The parasites of genus Leishmania are the causative agents of one of the most widespread and devastating diseases. According to follow-up data, these medications may provoke adverse drug reactions, drug resistance, relapse as well as financial burden. The mechanism of action of opioid drugs are primarily exerted via transmembrane G-protein coupled receptors. One of the potent synthetic immunomo...
متن کاملCluster of differentiation (CD) markers in erythrodermic patients: A case series study
Background: Erythroderma is an inflammatory disorder. It has various differential diagnoses, among which one of the most important is mycosis fungoides. Erythroderma itself can be a challenging disorder. Diagnosis of a mycosis fungoides patient presenting with erythroderma specially requires a careful assessment of the peripheral blood. Studies such as CD markers can lead to a more accura...

متن کاملPeripheral blood "endothelial progenitor cells" are derived from monocyte/macrophages and secrete angiogenic growth factors.
BACKGROUND Endothelial progenitor cells (EPCs) have been isolated from peripheral blood and can enhance angiogenesis after infusion into host animals. It is not known whether the proangiogenic effects are a result of such events as endothelial differentiation and subsequent proliferation of EPCs or secondary to secretion of angiogenic growth factors. METHODS AND RESULTS Human EPCs were isolat...
